Bacterial Staining: The Difference Between Microbial Extract Staining And Bacterial Staining

Microorganism

In recent years, the demand for non-toxic and non-toxic natural plant pigments has been increasing year by year. Because of the wide distribution and variety of microorganisms, microbial dyeing has a broad application prospect in the textile field.

Microbial pigments

Microbial pigment is a secondary metabolite of microorganisms. There are many kinds of pigments, including red, orange, yellow, green, green, green, purple, black, brown and other colors. Microbial pigments can be divided into water-soluble and non-water-soluble pigments. Compared with other natural dyes, the production cycle of microbial pigment is shorter, the cost is lower, and it is easier to industrial production.

There are two main ways to produce microbial pigment, one is secretion in the process of microbial growth, the other is a pigment formed by transforming a certain component in the culture medium as substrate. For the latter, it is necessary to add substances needed for pigment production in the culture medium to promote pigment production and increase pigment yield.

Microbial staining method

[extraction staining method]

Extraction dyeing method is to use liquid culture medium to cultivate microorganisms to metabolize a large number of pigments, and through the process of separation, extraction and concentration, the pigment solution is obtained.

The pigment solution can be used not only as dye solution, but also as pigment powder. The advantages of extraction dyeing method are wide range of application, easy to industrial production, but the disadvantages are tedious extraction process and high cost.

[cell staining method]

There are two methods of cell staining because of different culture medium. In the culture, when a large number of bacteria are put into the culture liquid to metabolize a large amount of dye; The other is solid agar medium. After a period of culture, when the microorganism metabolizes a large amount of pigment, water is added into the bacteria and medium, boiled, and then the fabric is dyed at 80 ℃.

The advantage of the method is that it is simple, time-saving and easy to operate. However, it is not suitable for microorganisms producing insoluble pigments.
